Two plain folders and sealed sample tubes for comparing materials

Identify the level of disagreement

Start by describing the difference precisely: participant identifiers, marker results, kinship index or verbal conclusion. These are four different tasks. Two documents may contain different percentages while supporting the same relationship in the same way. Conversely, similar final statements do not remove a discrepancy in original genetic observations concerning the same person.

For comparison, gather both complete reports, appendices and document version information. A working table states the date, provider, examination purpose, participants, objects and questions posed. A corrected report is considered with an explanation of changes: comparing an initial file with a final version without its history can create an artificial contradiction.

Match participants and samples

For each participant, trace the connection between the sampling document, packaging label, registration identifier and results table. Letters and numbers may differ between organisations; a clear mapping is needed rather than identical characters. Also check whether samples were collected anew or both examinations used material from the same initial source.

This distinguishes independence of laboratory procedures from independence of the material source. Reanalysing part of the same sample does not recheck whose material it was when obtained. At the same time, a documentary gap does not prove substitution: the reviewer establishes whether the connection can be restored and how the gap affects a particular answer. An assumption of intentional falsification requires separate grounds.

Compare genetic observations

First identify shared markers and check whether their designations and result presentation are comparable. Different test panels may provide different amounts of information. A locus missing from one document is not an allele mismatch. Different recording formats require an explanation from an appropriate specialist rather than mechanical equality checking of rows.

If a comparable marker genuinely has different reported results for one participant, profile quality and completeness, repeat analysis information and possible object limitations are examined. A substantive assessment may require original analytical records and laboratory explanations. A review of the final table alone cannot establish what signal the instrument observed or how the procedure was actually performed.

Align the calculations to the same question

When genetic data are the same, differences in indices are sought in the hypotheses compared, participants included, population frequencies, adjustments and treatment of mutations. It is important to establish whether the same relationship is compared with the same alternative. A calculation against an unrelated-person hypothesis is not interchangeable with one against a close-relative hypothesis.

The conversion to probability is checked separately: the prior assumption and rounding. Reproducing the statistics requires the algorithm, parameters and sufficient input data; where software is used, its version and settings may matter. If information is missing, the reviewer formulates a request rather than declares the higher percentage correct. A greater number of markers is also assessed by their informativeness for the task posed.

How comparison results are documented

Each difference is described by its source and significance for the conclusion. It is useful to distinguish explained format or rounding differences, differences in the task posed and unresolved data contradictions. For the latter, identify which document or examination may help and what it will not check. A new laboratory examination requires a separate brief and proper sample collection.

The outcome may reconcile two conclusions, identify an issue with a particular calculation or show that the submitted set does not allow a choice between hypotheses. A review does not create a new profile or promise to refute either document. At the organisational stage, it is enough to state the nature of the disagreement; transfer of complete genetic materials to the appropriate specialist is agreed through a suitable channel, without uploading identifying data through the general form.
